Techniques

Ion Channel Engineering

The techniques used include patch-clamp electrophysiology (recording of macroscopic and microscopic currents), molecular biology, site-directed mutagenesis, biochemistry, protein chemistry, protein expression and purification, cell transfection, cell culture, optical microscopy, fluorescent dye uptake, data analysis, and molecular visualization of proteins.

BioFunctional Chemistry

Synthetic chemistry, protein conjugation, microfluidic analysis, cell culture and expression of recombinant proteins.

Smart NanoParticules

  • Chemistry: multi-step organic synthesis, purification by chromatography (normal and reverse phase, preparative HPLC), characterization of organic molecules by NMR/mass spectrometry/HPLC analysis (diode array).
  • Physicochemistry: Dynamic light scattering (DLS), Nanoparticle tracking analysis (NTA), Rheology.
  • Spectroscopy: UV/visible spectrophotometry, spectrofluorimetry, luminometry, photochemical and/or photophysical characterizations (at 1 and 2 photons) of fluorophores, photosensitizers and photolabile groups.
  • Biomaging: confocal laser scanning microscopy, transmission electron microscopy (TEM).
  • Cellular and molecular biology: cell culture, cell viability/death tests, flow cytometry, protein and immunoenzymatic assays, gel electrophoresis (nucleic acids/proteins), immunocytochemistry, western blots.
  • In vivo biology: mouse model of acute lung inflammation or asthma, histology, immunohistochemistry.

Chemistry of Photoresponsive Systems

The activity of the PCS team is based on three major axes: chemistry, spectroscopy and bioimaging. We are using multiple techniques:

  • Chemistry: Multi-step organic synthesis, purification by chromatography (normal and reverse phase), and analysis by NMR/HPLC/mass spectrometry.
  • Spectroscopy: UV/visible/near-infrared spectrophotometry, spectrofluorimetry, high-power laser-excited spectroscopy. Single molecule microscopy, gel electrophoresis with fluorescence detection, multi-well plate reader (absorbance/fluorescence).
  • Bioimaging: Wide-field microscopy, laser scanning confocal microscopy, super-resolution microscopy based on single molecule detection.
